Information on genetic diversity in pummelo
germplasm is necessary to support breeding programs and efforts of
conservation. The objectives of this research were to know genetic diversity of
pummelo germplasm collected from Magetan based on morphological and isozyme
analyses and to know their phylogenetic. The research was conducted in Bendo
and Sukomoro, Magetan from April to September 2005 by exploration and continued
with isozyme assays in Laboratory of Molecular Biology, MIPA, Brawijaya
University.
Electrophoresis were chosen as isozyme observed Esterase (EST) and Aspartate aminotransferase (AAT) enzymes. Exploration resulted seven cultivars known as landrace in Magetan i.e., Gulung; Duku; Sri Nyonya; Adas Biasa; Adas Nambangan; Bali Merah and Bali Putih. Adas Nambangan had two different morphological characters namely, Nambangan A and Nambangan B. Genetic variabilities was detected on morphological traits, EST and AAT enzymes. Isozymes showed 9 to 12 banding patterns. Based on cluster analyses Adas Nambangan A has related to Adas Duku, Adas Nambangan B to Adas Biasa, Bali Merah to Bali Putih, and Gulung and Sri Nyonya to Adas Nambangan B and Adas Biasa.
